- a test for folic acid or vitamin B12 deficiency. A dose of the amino acid histidine, which requires the presence of folic acid or vitamin B12 for its complete breakdown, is given by mouth. In the absence of these vitamins, formiminoglutamic acid (figlu) - an intermediate product in histidine metabolism - accumulates and can be detected in the urine.
